What Is Bacitracin?

Cas number: 1405-87-4
Molecular formula: C66H102N17O16S
Molecular weight: 1421.68538
Bacitracin is a white to off-white fluffy powder. It is soluble in water, ethanol, and pyridine.
It is one of our China GMP-Approved APIs along with Bacitracin zinc Powder, polymyxin b sulfate Powder.
Its acidic or neutral aqueous solution is relatively stable.
When pH maintains at 5-7, its solution is stable for 4 weeks. When solution pH is greater than 9, bacitracin could degrade rapidly at room temperature.
First isolated in 1945 and later commercially produced, bacitracin is a mixture of related polypeptides widely used as an antibiotic in injections or topical formulations.
It is a mixture of related compounds with different antibacterial activities.
Related components include compounds A, A1, B, B1, B2, C, D, E, F, G, and X.
Of all those 11 components, compound A was found to have the most antibacterial activity.
Compound B1 and B2 have similar efficacies and are approximately 90% as active as compound A.
The rest of the components do not appear to be extensively studied.
it is an effective narrow-spectrum polypeptide with an antibacterial spectrum similar to penicillin and effective against Gram-positive bacteria, some Gram-negative bacteria, spirochetes, and actinomycetes.
These polypeptides inhibit Gram-positive bacteria by interfering with the cell wall and peptidoglycan synthesis.
What Are Bacitracin Related Ingredients?
As per Monographs in USP38-NF33 S2, it has 8 related ingredients:
A, B1, B2, B3, C1, C2, C3, and F, are the impurities listed in USP.
But according to Wikipedia, it is made up of 11 components: A, A1, B, B1, B2, C, D, E, F, G, and X.

Among the 8 substances, 4 of them are active ingredients: A, B1, B2, and B3.
The rest of the ingredients are regarded as impurities such as Bacitracin C1, C2, C3, and F.
How Is Bulk Bacitracin Powder Produced?
It is commercially manufactured by growing the bacteria Bacillus subtilis var Tracy I in a container of liquid growth medium.
After a certain period, the bacteria synthesize the antibiotic and secrete the antibiotic into the medium.
Then the antibiotic is extracted from the medium through chemical processes.
1. Fermentation:
Use Bacillus subtilis and B. licheniformis to get its solution.
The solution is an extraction and separation solution of a Bacillus subtilis and a B-type Bacillus licheniformis fermentation broth with a potency of no less than 5000 u/ml and conductance of less than 2000 μs/cm.
2. Adjusting PH value:
An alkaline solution is added to its solution to adjust the pH from 8 to 11.
Adjust pH from 5 to 7 with an inorganic acid.
Stir it for 1 to 2 hours, and let it stand for 5 to 10 hours.
3. Extraction and Purification:
it is then obtained by membrane separation, drying, pulverization, and sieving.
The spray-dried powder obtained by processing above can be used as pharmaceutical raw materials for medicinal preparations such as injections and ointments with polymyxin b.
USP requires that the total content of compound A is no less than 40.0%.
The total amount of A, B1. B2 and B3 should be greater than 70.0% and the potency is no less than 60IU/MG.
What Is Bacitracin Powder Used For?
It is used for manufacturing human medicines as a polypeptide antibiotic against infections.
Please note, that it should only be used to treat or prevent infections that are proven or strongly suspected to be caused by susceptible bacteria.
It is primarily made into ointments as it is can cause kidney damage when it is taken orally.
It has been indicated in topical formulations for acute and chronic localized skin infections.
It is also used intramuscularly for infantile streptococcal pneumonia and empyema.
It is also formulated as an ointment with neomycin and polymyxin B as OTC.
Its ointment formulated with neomycin and polymyxin B along with hydrocortisone is indicated for the treatment of corticosteroid-responsive dermatoses with secondary infection.
People also use it as an aftercare antibiotic on tattoos and circumcision.
Bacitracin is preferred over combination products such as Neosporin because of its fewer ingredients, which lowers the chances of an allergic reaction.
Although its allergic cross-reaction with sulfa drugs has been occasionally reported, bacitracin-containing topical preparations are a great alternative to silver sulfadiazine (Silvadene) for patients burnt with a sulfa allergy.
In the biochemical industry, researchers would use it to distinguish Streptococcus pyogenes from other “strep” bacteria, with S. pyogenes being sensitive to bacitracin and others resistant.
It is also used to detect Haemophilus influenza colonies amongst respiratory flora.
Because H. influenza is intrinsically resistant to it, colonies would form within the zone of inhibition.
Bacitracin vs Mupirocin
#1 Different Usages
Mupirocin is an antibacterial pseudomonic acid antibiotic produced by Pseudomonas fluorescens.
It is mainly used for skin infections such as impetigo, other primary or secondary superficial skin infections, such as ecthyma, eczema, folliculitis, furunculosis, atopic dermatitis, epidermolysis bullosa, and minor wounds, burns, or ulcers.
While bacitracin is used for infection prevention, especially for wounds on the skin such as cuts or burns.
It is also used for infantile streptococcal pneumonia and empyema when used intramuscularly.
#2. Different working mechanism
Mupirocin works by preventing bacteria from growing.
It inhibits protein synthesis in bacteria by binding to isoleucyl transfer RNA synthetase.
While bacitracin is a powerful bacteria killer so it will prevent wounds from infections.
#3 Different Dosage forms
There are mainly 4 dosage forms of Mupirocin:
1. Mupirocin Calcium Ointment
2. Mupirocin Cream
3. Mupirocin Nasal Ointment
4. Mupirocin Ointment.
Dosage forms for bacitracin include:
1. Injections:
2. Ointment for skin
3. Ophthalmic ointment.
As we can there are only topical dosage forms for mupirocin, but without injections.
Neither does Mupirocin have eye ointments, while bacitracin has injections as well as ophthalmic ointments.
The reason that mupirocin can not be made into eye ointments is that: once mupirocin is applied to the eyes, there will be severe symptoms such as burning and tearing.
It will take days or even weeks for this uncomfortableness to disappear. (If contact with the eyes occurs, thoroughly wash eyes with water or go to a doctor immediately).
However, there is mupirocin ointment for intranasal use, while there are no bacitracin dosage forms suitable for intranasal uses.
#4 Different Costs
The unit price for mupirocin API is around 2000usd per kg, almost 3 times that of bacitracin.
It is the same case when they are made into formulations.
Mupirocin formulations are also more expensive than bacitracin.
Check the price below:
the lowest Bactroban price is 320.38usd and the lowest price for bacitracin ointment is 50.83usd.
Conclusion: Mupirocin is more expensive and used for infections.
Bacitracin is cheaper and used for infection prevention.
Mupirocin can not be used for eye infections but has no problems with nose infections.
Bacitracin has injections and can be used for infections of your eyes.

Bacitracin vs Neosporin
Differences between those two are as below:
#1 Number of active pharmaceutical ingredients
There are 3 active ingredients in Neosporin: neomycin sulfate, bacitracin zinc, and polymyxin b sulfate.
While bacitracin ointment only contains bacitracin.
#2 Chances of allergy
More people are allergic to Neosporin while no problem with bacitracin ointment.
That’s because Neosporin contains more active ingredients than bacitracin ointment, especially when neomycin within is prone to cause allergies.
When people are allergic to neomycin sulfate and polymyxin b sulfate, they can only use bacitracin ointment for another try.
Bacitracin has less chance to give you allergies.
If you are allergic to bacitracin, you must be allergic to Neosporin, that’s for sure.
#3 Bacteria Killing Range
Neosporin is a combination of antibiotics, so it has a wider bacteria-killing range and a better infection-preventing effect than bacitracin.
#4 Production process
Because bacitracin ointment only contains one active ingredient, the production process is relatively easier.
Trust me, mixing all the 3 ingredients in Neosporin evenly is not an easy job.
It takes longer to blend and the production process is more complicated.
#5 Price
As Neosporin is a famous brand, it costs more than bacitracin ointment in many markets.
Even when you compare Neosporin with Triple Antibiotics containing the same active ingredients as Neosporin, it still costs more.
What Is The HS Code Of Bacitracin?
It belongs to the class of organic compounds known as polypeptides.
These are peptides containing ten or more amino acid residues.
It is classified as other antibiotics with HS Code 2941909090 in China Customs.
If you are importing its finished dosages such as ointments, injections, related hs code should be 3004909090.
How Much Does Bulk Bacitracin Powder Cost?
For commercial order quantities, the price is about 750usd/kg as Fob Shanghai/Beijing.
If you only need several kilos of non-sterile powder for your pharmacy compounding or lab research, our price is around 1100usd/kg for your ref.
In case you need it by 10kg for pilot manufacturing of ointments, our offer is about 950usd/kg.

How To Ship and Store Bacitracin Powder?
#1. Shipment
First of all, it is a very safe chemical to be shipped by air.
It has been tested by DGM and regarded as a common and safe chemical since 2008 or earlier.
And it is suitable for shipment by air, by express and by sea packed in normal packing materials
Certificate of safe air transportation issued by DGM has shown that bacitracin is non-flammable, not dangerous, not poisonous, etc.
As to how you want to ship the pure powder, it all depends on its quality standards.
If you are buying injection-grade powder, you may want it to be shipped as fast as you can at a low temperature.
Shipping by air or by express is your best option, to rule out any possible high temperatures.
Because the injection-grade powder is more expensive than oral-grade types, you have good reasons to spend extra bucks on the shipping for it.
If you are procuring some bulk powder for research such as bacterial growth, you may ship it by express/courier, with or without ice pads as you like.
Though normal-temperature shipment will not affect its activity too much, it is better you ship it at a low temperature to make sure its potency.
#2. Storage
The bulk powder is relatively stable during storage.
Our stability tests have shown that it is stable for at least 36 months when it is stored in a dry environment and protected from the sunlight at room temperature.
So you can store the powder according to the conditions above if you are storing it for topical formulations.
For storage of injection-grade powder, low-temperature storage conditions are recommended.
