What Is Bacitracin Zinc?

Bacitracin zinc is a complex of zinc and bacitracin. It is a white or off-white powder with its characteristic odor and bitter taste, a mixture of related cyclic peptides produced by organisms of the licheniformis group of Bacillus subtilis var Tracy. These peptides disrupt Gram-positive bacteria by interfering with the cell wall and peptidoglycan synthesis
It is one of our USFDA-Approved pharmaceutical ingredients along with bacitracin and polymyxin b sulfate. It is an effective narrow-spectrum antibiotic peptide with a similar antibacterial spectrum to penicillin, mainly for Gram-positive cocci and bacilli. Zinc bacitracin powder also exerts a certain inhibitory effect on several Gram-negative bacteria, spirochetes, and actinomycetes with attributes of high efficiency, low toxicity, and low residue.
First thing first, we must mention it upfront, there are two types of powder: 1. high-potency type Pharm-Grade (Potency No Less Than 65IU/MG); 2. Feed-Grade or Industrial-Grade (Potency Less than 40IU/MG). Here we mainly discuss pharm-grade type.
How Is Bacitracin Zinc Powder Produced?
1. Fermentation process using Bacillus subtilis and B. licheniformis to get bacitracin solution. (Bacitracin solution is an extraction and separation solution of a Bacillus subtilis and a B-type Bacillus licheniformis fermentation broth with a potency of no less than 5000 u/ml and a conductance of less than 2000 μs/cm).
2. An alkaline solution is added to the bacitracin solution to adjust the pH as 8 to 11, add inorganic zinc salt, adjust pH between 5 to 7 with an inorganic acid, stir for 1 to 2 hours, and let stand for 5 to 10 hours. (The alkaline solution can be either ammonia water or sodium hydroxide. The inorganic zinc salt is one of zinc oxide, zinc chloride, and zinc sulfate. The inorganic acid is one of hydrochloric acid and sulfuric acid.)
3. It is then obtained by separation, drying, pulverization, sieving, inspection.
The total content of bacitracin A should be greater than 40.0%, the total amount of bacitracin A, B” B2 and B3 should be greater than 70.0%, Potency should be no less than 65IU/MG.
Zinc Bacitracin obtained by this method could be used for medicinal preparations such as aerosol sprays and ointments.
What Can Bacitracin Zinc Powder Be Used For?
It can be used by combining with other topical antibiotics (usually Polymyxin B sulfate and Neomycin) as an ointment (well-known as Pfizer’s Neosporin). It is used for the topical treatment of a variety of localized skin infections, as well as for the prevention of wound infections.
It is also indicated for Empyema, Infection NOS, Pneumonia, Corticosteroid-responsive Dermatosis, Infection in minor cuts, scrapes, or burns, Inflammatory ocular disease, etc (Made into cortisporin ophthalmic ointment).
Its ointments are used as first aid to help prevent infection in minor cuts, scrapes, burns.
There are people using its formulations for acne when acne pimples are infected. People would combine its ointment with cleansers containing benzoyl peroxide to kill bacteria that might colonize an injured pimple.
However, the method above does not always cure acne. Some people reported no improvement or even worse outbreaks after applying it.
There are ophthalmic solutions of non-ointment forms for eye infections. There are also Ophthalmic Ointments with Polymyxin B Sulfate indicated for the treatment of superficial infections of the external eye and its adnexa caused by susceptible bacteria.
What Are Bacitracin Zinc Dosages?

Topical Ointment, Strength: 500IU/G [Each gram contains Bacitracin Zinc (equal to 500 bacitracin units)]
Ophthalmic Ointment of 3.5 grams in tubes. Each gram equivalent to 500 bacitracin units, polymyxin B sulfate equivalent to 10,000 polymyxin B units, mineral oil, and white petrolatum.

Bacitracin zinc and bacitracin are produced in the same way but processed differently to get different products. Zinc within bacitracin makes it more stable. BeLow we list the differences between the two.
As we mentioned above, zinc salt makes the peptide bacitracin more stable when exposed to air and moisture. Though they have the same shelf life as three years when properly stored and sealed in our certificate of analysis, it will be another case when the package is broken.
Because bacitracin is very easy to absorb moisture and decompose, when the highest bacitracin concentration is reached in the fermentation process, we add zinc salt directly to culture solution to get a more stable zinc complex, namely zinc bacitracin. Bacitracin zinc is widely used in the feeding industry because of its inherent characteristics and is currently the most used antibiotic feed additive. Compared with bacitracin, the stability of bacitracin zinc is extremely superior: in a dry state, its potency can be kept at room temperature for 3 years without any change, even in acidic, alkaline or hot environments, its stability is also better. According to reports, when bacitracin zinc is stored for 24 months at a temperature of 40°C, its potency is only lost by 10%; however, bacitracin in feces begins to decompose at room temperature in 3 to 6 days, and it decomposes in compost even faster

Market demand for bacitracin zinc is much bigger than bacitracin. On one hand, bacitracin zinc is used in more ointments and aerosol sprays than bacitracin. Take the famous brands such as Neosporin and Polysporin for example, they both contain bacitracin zinc and polymyxin b sulfate, but neosporin contains the extra third ingredient neomycin sulfate. Only very small portion of the people who are allergic to neomycin sulfate would have to choose ointments containing bacitracin only.
