Trilostane is a white to off-white crystalline powder, marketed under Vetoryl for veterinary uses.

It is soluble in DMSO and DMF but difficult to be dissolved in water, with a solubility at 20°C is of about 59.3mg/l.

It used to be approved for the treatment of Cushing’s syndrome for humans until it was withdrawn in 1994 from the USA.

Currently, it is mainly used for Cushing’s in dogs and cats (pituitary-dependent hyperadrenocorticism and adrenal-dependent hyperadrenocorticism).

It was also used for breast cancer in the UK.

Some compounding pharmacies also have it compounded for the skin disorder Alopecia X for dogs.

Please note that it is not allowed to give pets trilostane with hepatic or renal diseases.

Avoid using it simultaneously with other drugs such as mitotane and ketoconazole that suppress adrenal activities.

What Is Trilostane Used For?

Trilostane is a medication that is used to treat a variety of conditions in both humans and animals.

In humans, it is primarily used to treat Cushing’s disease, a hormonal disorder caused by excessive production of the hormone cortisol.

In animals, it is used to treat Cushing’s disease as well as other hormonal disorders such as hyperadrenocorticism.

Cushing’s disease is a condition that occurs when the body produces too much cortisol, a hormone that helps regulate metabolism, immune function, and stress response.

High levels of cortisol can lead to a range of symptoms including weight gain, fatigue, high blood pressure, and thinning skin.

Trilostane works by inhibiting the enzyme that is responsible for the production of cortisol, thereby reducing the levels of cortisol in the body.

In addition to its use in the treatment of Cushing’s disease, trilostane has also been used to treat other hormonal conditions such as hyperadrenocorticism in animals.

Hyperadrenocorticism, also known as “Cushing’s syndrome,” is a condition that occurs when the adrenal glands produce too much cortisol.

This can lead to a range of symptoms including weight gain, thinning skin, and muscle weakness.

Trilostane is effective in reducing the levels of cortisol in the body and can help to improve symptoms in affected animals.

Trilostane is usually taken orally in the form of a capsule or tablet.

The dosage and frequency of treatment will depend on the specific condition being treated and the individual patient.

It is important to follow the instructions of a healthcare provider or veterinarian when taking trilostane to ensure the best possible results.

It is important to report any side effects to a healthcare provider or veterinarian as they may need to adjust the dosage or frequency of treatment.

it can cause some side effects in some individuals and it is important to report any side effects to a healthcare provider or veterinarian.

What Are Trilostane Side effects?

Trilostane is a fast-acting agent with very few side effects.

While it is well-tolerated for most cats and dogs, you need to watch your pets closely after they are medicated.

Side effects include but are not limited to: loss of appetite, vomiting, diarrhea, lethargy, unstable walking, weakness, heavy panting, muscle pain, and excessive drooling.

You can try to avoid those side effects from the start by using smaller dosing than recommended dosages and later increasing the dosing slowly to recommended dosages.

In case of serious conditions such as non-stopping vomiting, or choking, contact your veterinarian immediately.

How To Feed Trilostane To Cats And Dogs?

When our dogs or cats are sick and need to take trilostane for their sufferings, how do feed them?

#1. How to feed

Due to the primary side effect of losing appetite, it is recommended to stash the capsule into their favorite foods, but do not give them too much, as they may throw up later from being too full.

Try to give them the medication on a routine schedule, once daily, at the same time of the day, and preferably in the morning with an empty stomach.

When you are stashing the branded capsules or compounded ones, make sure you are wearing disposable gloves for protection.

If you do not have any gloves around, make sure you wash your handly thoroughly after handling them.

Take extra caution especially are you planning to be pregnant.

Do not break a capsule to make up for proper dosing.

#2. Daily dosage

To prevent possible side effects, your veterinarian would advise you to start with the lowest dosing according to body weight.

The treatment dosing of hyperadrenocorticism for dogs is 2.2mg~ 6.6mg/kg.

There are capsules of 5mg, 10mg, 30mg, 60mg, and 120mg available, make your combination after your calculation.

After 10-14 days of medication, take your pets to your veterinarian for a re-examination, no matter whether it is good or bad. Follow your vet’s advice on how to proceed further.

How To Store Trilostane Powder?

In case you have bought or imported more powder than you can use for one compounding or production, you will need to store them for future use, how you should store them to keep them fresh as a new batch?

Below is how:

Make sure your powder is packed in an air-tight, well-sealed foil bag, protected from heat, moisture, and light.

If you have vacuum packing equipment, it can not be better with vacuum packing.

The shelf life for the pure powder is at least 2 years at a room temperature of 25°C.

If the temperature is too high such as 35 or 40 degrees, it will make its shelf life shorter.

If it is stored in a freezer, its shelf life will be over 4 years.

No matter how long it has been stored, to make sure of its quality and medical efficacy, retest its purity by HPLC and structure by 1HNMR before you plan to use it again during storage.

Though it is sensitive to temperature fluctuations, a small and mild change is allowed for its storage (20°C~30°C).

As long as the temperature change is not dramatic, it will be fine.

Just make sure it is not 30°C in one minute and 20°C in another.

If you only have a small quantity of its powder, feel free to store it under -20°C in your refrigerator.

Seal it tightly to avoid any moisture.
